Tripeptide-1, its salts and the copper complex
Tripeptide-1 is the Gly-His-Lys peptide, commonly abbreviated GHK. CosIng describes Tripeptide-1 Acetate separately as the acetate salt, and Copper Tripeptide-1 as a copper complex of the peptide. For a GHK enquiry, name the required form rather than using Tripeptide-1 and GHK-Cu as interchangeable descriptions.
A salt requirement belongs in the purchase description together with the peptide identity and intended cosmetic use. Ask for the counterion and its content where relevant, then check that the stated formula and assay basis correspond to that form. The free peptide and a salt can carry different formula weights, so a peptide name alone is insufficient for a mass-based formulation calculation.
Please tell us whether your project requires free GHK or a specified salt so we can confirm the form proposed for quotation. Requests for the copper complex belong with our separate GHK-Cu product entry.
Peptide purity and GHK content are separate questions
For Tripeptide-1, request identity evidence for the Gly-His-Lys sequence together with the quantitative acceptance test. Ask whether a reported purity represents chromatographic peak area or a measured peptide content in the supplied material. Include the calculation basis and the reference standard used for the GHK result in your document request.
A salt or preparation may also contain counterions, water or other constituents that need their own description. Specify the relevant water, residual solvent and related-peptide limits for your cosmetic raw-material assessment. For impurity review, request analytical information appropriate to the proposed synthesis and purification route without assuming that every GHK source has the same profile.
The SCCS guidance asks for the specific chemical form and representative material when assessing a cosmetic ingredient and its derivatives. Use the specification, composition statement and any proposed COA together to check that they all describe the same Tripeptide-1 form.
Using the GHK form in a cosmetic development brief
The CIR review describes skin conditioning as a principal cosmetic role of the peptide group that includes Tripeptide-1. That review treats GHK, its copper complex and fatty-acyl derivatives as distinct ingredients. Published information for one derivative should therefore be checked for relevance before it is used to assess another material.
For your GHK formulation, include the target concentration, solvent system and expected processing conditions in the enquiry. Ask for dissolution data tied to the specified form and test temperature, and evaluate it in your own base. If you need a liquid preparation, request the peptide concentration and all accompanying ingredients so your formulator can calculate additions correctly.
Check stability over your planned processing and storage conditions using the proposed GHK material. Request packaging, retest or expiry information and the available supporting documents for that composition before agreeing an order.
Product identity
The applicable INCI name depends on the supplied form: the free base is listed as TRIPEPTIDE-1, the acetate as TRIPEPTIDE-1 ACETATE. Tell us the form you need and we will confirm the INCI name for it.
CAS and formula (verified against PubChem)
| Form | CAS No. | Molecular formula | Molecular weight | PubChem CID |
|---|---|---|---|---|
| acetate | 72957-37-0 | C16H28N6O6 | 400.43 | 22214678 |
| free base | 49557-75-7 | C14H24N6O4 | 340.38 | 73587 |
Molecular formula and weight describe the substance identified by that CAS number. Confirmation that a specific batch matches this identity comes from the specification sheet and COA.
